Introduction

SIEVEWELL Slide is a device that has a self-standing filter membrane with ca. 370,000 nanowells. Each nanowell has microscopic through-pores connecting the liquid above the filter membrane to two side ports through a micro-gap situated below the filter membrane. When liquid is suctioned from the side ports, cells move along with liquid flow, and are trapped by hexagonal nanowells as single-cell. Once cell is trapped by the nanowell, chance for further entering of cells into the same nanowell should significantly decrease by diminished liquid flow around the nanowell caused by (partial) clogging of the through-pores with the trapped cell. This would give high single cell rate (more than 95% for HL60, PC-3, A549, and MCF-7 cancer cell lines, at the number of 2×105 cells).

Slide Description

The Y04C plate has 4 independent units (A–D), each with 6
inlet wells (1–6), a cell inlet (8), and a large outlet well (7).
Each row of wells (A–D) addresses the corresponding culture
chamber. The plate is shipped preprimed with a PBS (phospatebuffered saline) solution, which can be replaced with a buffer
of choice prior to experiment. Each chamber has trap regions
of 3.5, 4.0, and 4.5 µm in height to hold cells in a single focal
plane during long-term analysis. The plate is for single use only.

Limitations

The slide is incompatible with acetone. Slides should be tested for compatibility with other acids or organic solvents prior to use.
